About the Course

This intermediate one-day workshop will provide the analytical scientist with a clearer understanding and solid working knowledge of best practices (method development, operation, and troubleshooting) of HPLC and UHPLC (ultra-high-pressure liquid chromatograph). The focus is on pharmaceutical analysis of small molecule drugs. A brief survey of biopharmaceutical, food, environmental, chemical, and bioscience applications is included.

  • HPLC Applications and an Overview of UHPLC
  • HPLC Method Development and Validation
  • HPLC Operation and Troubleshooting

Recommended textbook: M. W. Dong, HPLC and UHPLC for Practicing Scientists, 2nd Ed., Wiley, Hoboken, NJ, 2019

What You Will Learn

  • Concepts, perspectives, best practices, and potential issues of UHPLC
  • Fundamentals of HPLC method development and easier approaches
  • Overview of method validation and transfer
  • Standard operating procedures for HPLC modules, and troubleshooting strategies

Who Should Attend

Analysts, scientists, researchers, and managers in pharmaceutical and other industries who want to get an updated overview of the best practices of HPLC and UHPLC in pharmaceutical analysis including HPLC method development, operation, and troubleshooting. A basic understanding of chemistry and HPLC with some hands-on experience is assumed.

Course Outline

  • HPLC Applications and an Overview of UHPLC

    A.      Overview and perspectives, UHPLC concepts (the need for high pressure, void volume, peak volume, instrumental bandwidth), UV flow cell design, dwell volume

Benefits and case studies: very fast separations with good resolution, very high-resolution analysis of complex samples, facilitating rapid method development, flexibility to customize resolution

    B.      Potential issues and how to mitigate (viscous heating, operating nuances, compatibility to existing methods, injection precision, detector sensitivity vs. mixing volumes), method translation from HPLC to UHPLC and vice versa using geometric scaling, how to transition from HPLC to UHPLC, end-fittings

    C.      HPLC applications including pharmaceutical, biopharmaceuticals, food, environmental, chemical, and bioseparations

    E.      UHPLC overview, perspectives, and benefits

    F.      Potential issues and how to mitigate

  • HPLC Method Development and Validation

    A.    Tradition strategy and a survey of automated tools

    B.    3-pronged template approach

    C.    Use of a universal generic gradient method

    D.    Method validation and method transfer

  • HPLC Operation and Troubleshooting

    A. Mobile phase and sample preparation and best practice in HPLC operation

    B. Problem diagnosis and troubleshooting guide with case studies

    C. Diagnosing and solving problems (pressure, baseline, peak, data performance)

    D.    Mobile phase and sample preparation and HPLC operation

    E.    Maintenance and troubleshooting guide

    F.   Diagnosing and solving problems
